      <title>Double pH- along with Temperature-Responsive Necessary protein Nanoparticles.</title>

We previously described a liquid culture material, LA717, which prevents nonclonal cell adhesion and subsequent clumping, thus allowing clonal growth of spheroids in an anchorage-independent manner. Here, we examined such liquid culture cancer spheroids for the acquisition of apical-basal polarity, sensitivity to an Akt inhibitor (anticancer drug MK-2206) and interaction with ECM. The spheroids present apical plasma membrane on the surface, which originated from the failure of polarisation at the single-cell stage and subsequent defects in phosphorylated ezrin accumulation at the cell boundary during the first cleavage, failing internal lumen formation. At the multicellular stage, liquid culture spheroids presented bleb-like protrusion on the surface, which was enhanced by the activation of the PI3K/Akt pathway and reduced by PI3K/Akt inhibitors. Liquid culture spheroids exhibited slow proliferation speed and low endogenous pAkt levels compared with gel-cultured spheroids and 2D-cultured cells, explaining the susceptibility to the Akt-inhibiting anticancer drug.       <title>Anti-C1q antibody inside people with lupus nephritic flare: 18-month follow-up plus a stacked case-control review.</title>

Colloidal systems prepared from carbohydrates are subject of intense research due to their potential to enhance drug permeability through biological membranes, however their characteristics and performance are never compared directly. Here we report the results of a comparative investigation of a series of butylglyceryl-modified polysaccharides (chitosan, guar gum, and pullulan) that were formulated into nanoparticles and loaded with a range of model actives (Doxorubicin, Rhodamine B, Angiotensin II). Butylglyceryl-modified guar gum and corresponding pullulan nanocarriers were more stable at physiological pH compared to those obtained from modified chitosan, and studies of the in-vitro interactions with mouse brain endothelial cells (bEnd3) indicated an increased biological membrane permeability and lack of toxicity at application-relevant concentrations. No significant haemolytic effect was observed, and confocal microscopy and flow cytometry studies confirmed the efficient cellular uptake and cytoplasmic localisation of NPs. Most promising characteristics for brain drug delivery applications were demonstrated by butylglyceryl pullulan nanocarriers. A low-molecular-weight chitosan (LMWC) sample was prepared by enzymatic hydrolysis, and used for investigation of special Maillard reaction products (MRPs) and factors affecting LMWC bioactivities. After undergoing MR, LMWC turned to brown color (termed BLMWC), showed reduction of several indices of rice growth promotion. This alteration of bioactivities was attributable to MRPs in BLMWC. A special MRP, 5-hydroxy-2-pyridine methanol isomer (5-H-2PMIS), was identified by HPLC and LC-MS. Analysis of key factors affecting MR, using this MRP as monitoring target compound and OD420 value, suggested that MR process can be minimized by storing LMWC under vacuum in a dry, low-temperature, neutral-pH environment. Na2SO3 was effective for inhibition of MR, at optimal concentration 0.5 %. Chemical and FTIR analyses showed that Na2SO3-treated sample conformed to the Chinese National Standard of chitosan (GB 29941-2013). Control of MR is essential for application of LMWC in food, pharmaceutical, and other industries. Carrageenan fibers have attractive applications in textile, but their low strength remains a problem that needs to be urgently addressed. In this work, a novel facile, environmental friendly method for fabricate high-strength carrageenan fibers is proposed. It involves the crosslinking of a small amount of Ba2+ ions in the carrageenan solution, followed by using recyclable alcohol in coagulation and stretching baths. Carrageenan molecular chains were allowed to first sufficiently interact with metal barium ions, and then were stretched and dehydrated with alcohol to increase the hydrogen bonding interaction between the molecular chains. As a result, the carrageenan fibers with high-strength ionic and hydrogen bonds were obtained. The fibers obtained by the novel method had high tensile strength at 1.63 cN/dtex, which is two times higher than that of those obtained by the traditional process. The aim of the present study is to characterize the structure of a novel natural polysaccharide from Agrocybe aegirita (AAPS) and evaluate its anti-aging activity. The MALLS and GC-MS analysis indicated that the AAPS with molecular weights of 1.81 × 104 Da was mainly composed by rhamnose, fucose, mannose, and glucose in a molar ratio of 2.9010.253.7038.27. The FT-IR and NMR analysis showed that the backbone of AAPS was α-L-Rhap-(1→6)-β-D-Glcp-(1→2)-α-L-Fucp-(1→6)-α-D-Glcp-(1→5)-α-L-Araf-(1→4)-β-D-GlcpA-(1→5)-α-L-Araf-(1→6)-α-D-Manp-(1→6)-α-D-Manp-(1→2)-α-L-Fucp-(1→6)-β-D-Glap-(1→2)-α-L-Rhap-(1→6)-β-D-Galp-(1→, which linked with two side chains α-L-Fucp-(1→6)-β-D-Glcp-(1→6)-β-D-Manp-(1→ and α-D-Xylp-(1→2)-α-L-Fucp-(1→5) -α-D-Araf-(1→6)-β-D-Galp-(1→ at OH2 at H-4-arabinose and the terminal Galp residues, respectively. There was no significant difference in the 30-day mortality \[primary group 7.4%; redo group 3.2%; 95% confidence interval (CI) (0.19-35.29); P = 0.63\] and the 3-year mortality \[primary group 22.2%; redo group 16.7%; 95% CI (0.23-3.23); P = 0.72\] between redo and primary cases. We explored their possible interplay in diabetes and associated complications. The variant allele and haplotype frequencies at these polymorphic loci vary among different super-populations (African, European, admixed Americans, South and East Asians). The variant alleles are particularly highly prevalent in different European and admixed American populations. Differential distribution of these variants in different ethnic groups suggests that certain drugs might be more effective in selective populations rather than all. Therefore, population specific genetic architectures should be considered before considering a drug for these conditions.We aimed to assess the clinicopathological features and to determine the prognostic factors of cervical adenocarcinoma (AC). Relevant data were extracted from surveillance, epidemiology and end results database from 2004 to 2015. The log-rank test and Cox proportional hazard analysis were subsequently utilized to identify independent prognostic factors. A total of 3102 patients were identified. The enrolled patients were characterized by higher proportion of early FIGO stage (stage I 65.9%; stage II 14.1%), low pathological grade (grade I/II 49.1%) and tumor size ≤ 4 cm (46.8%). The 5- and 10-year cancer-specific survival rates of these patients were 74.47% and 70.00%, respectively. Meanwhile, the 5- and 10-year overall survival (OS) rates were 71.52% and 65.17%, respectively. Multivariate analysis revealed that married status, surgery as well as chemotherapy were independent favorable prognostic indicators. Additionally, aged &gt; 45, tumor grade III/IV, tumor size &gt; 4 cm, advanced FIGO stage and pelvic lymph node metastasis (LNM) were unfavorable prognostic factors (all P  less then  0.01). Stratified analysis found that patients without surgery could significantly benefit from chemotherapy and radiotherapy. In addition, chemotherapy could significantly improve the survival in stage II-IV patients and radiotherapy could only improve the survival in stage III patients (all P  less then  0.01). Marital status, age, grade, tumor size, FIGO stage, surgery, pelvic LNM and chemotherapy were significantly associated with the prognosis of cervical AC.</p>

Independent of the ancestral contribution, two SNPs were associated with a reduced risk (p ≤ 0.01) of developing COPD in the CYP2A6 (rs4105144) and CYP2B6 (rs10426235) genes; however, a haplotype was associated with an increased risk of COPD (p = 0.007, OR = 2.47) in the CHRNA5-CHRNA3 loci among smokers with a high Caucasian component. The objective of the study was to assess the positive and negative investment of these types of sites based on the available information on the possibilities of investing in the implementation of the brownfield industrial park compared to the greenfield in the western part of East Slovak region and to identify a more appropriate alternative of investing. Based on the assessment of investment in the industrial parks, the appropriateness of the allocation of investment capital was assessed through the simple additive weighting (SAW) method. The SAW approach allows us to objectivize the weighting value of selected factors and thus assess the appropriateness of the allocation of investment capital. Based on the results, it is more advantageous to allocate the investment capital to the greenfield as the return on investment of the project expressed as a percentage of the average annual profit was 9.5%, compared to brownfield with only 2.9%.With the aim to fully exploit the by-products obtained after the industrial extraction of starch from sweet potatoes, a cascading approach was developed to extract high-value molecules, such as proteins and pectins, and to valorize the solid fraction, rich in starch and fibrous components. This fraction was used to prepare new biocomposites designed for food packaging applications. The sweet potato residue was added to poly(3-hydroxybutyrate-co-3-hydroxyvalerate) in various amounts up to 40 wt % by melt mixing, without any previous treatment. The composites are semicrystalline materials, characterized by thermal stability up to 260 °C. For the composites containing up to 10 wt % of residue, the tensile strength remains over 30 MPa and the strain stays over 3.2%. A homogeneous dispersion of the sweet potato waste into the bio-polymeric matrix was achieved but, despite the presence of hydrogen bond interactions between the components, a poor interfacial adhesion was detected. Considering the significant percentage of sweet potato waste used, the biocomposites obtained show a low economic and environmental impact, resulting in an interesting bio-alternative to the materials commonly used in the packaging industry. Thus, according to the principles of a circular economy, the preparation of the biocomposites closes the loop of the complete valorization of sweet potato products and by-products.A Fabry-Pérot acoustic sensor based on a graphene oxide membrane was developed with the aim to achieve a faster and simpler fabrication procedure when compared to similar graphene-based acoustic sensors. Unexpectedly, we find two types of ATP-Mg2+/Pi carriers, the canonical ones and shortened variants lacking the CaM-like domain. Phylogenetic analysis shows that both SCaMC variants have a common origin, unrelated to AACs, suggesting in turn that recurrent losses of the regulatory module have occurred in the different phyla. They are excluding variants that show a more limited distribution and less conservation than AACs. Interestingly, these truncated variants of SCaMC are found almost exclusively in parasitic protists, such as apicomplexans, kinetoplastides or animal-patogenic oomycetes, and in green algae, suggesting that its lost could be related to certain life-styles. Both Abs display comparably high affinities for distinct epitopes of viral Spike (S)-protein trimers. Gluc activity is reconstituted when the Abs are simultaneously bound to S-protein bringing the Ab-fused N- and C-terminal fragments close enough together (8 nm) to fold. We thus achieve high specificity both by requirement of simultaneous binding of the two Abs to the S-protein and also, in a steric configuration in which the two Gluc complementary fragments can fold and thus reconstitute catalytic activity. Gluc activity can also be reconstituted with virus-like particles that express surface S-protein with detectable signal over background within 5 min of incubation. Design principles presented here can be readily applied to develop reporters to virtually any protein with sufficient available structural details. Detailed clinical, imaging and biological characterisation of this population, along with the identification of mechanisms linked with disease progression and prognosis, would allow for tailored treatments and provide important mechanistic insights into the complex HFpEF pathophysiology.Recombinant antibodies (Abs) against the SARS-CoV-2 virus hold promise for treatment of COVID-19 and high sensitivity and specific diagnostic assays. Here, we report engineering principles and realization of a Protein-fragment Complementation Assay (PCA) detector of SARS-CoV-2 antigen by coupling two Abs to complementary N- and C-terminal fragments of the reporter enzyme Gaussia luciferase (Gluc). Both Abs display comparably high affinities for distinct epitopes of viral Spike (S)-protein trimers. Gluc activity is reconstituted when the Abs are simultaneously bound to S-protein bringing the Ab-fused N- and C-terminal fragments close enough together (8 nm) to fold. We thus achieve high specificity both by requirement of simultaneous binding of the two Abs to the S-protein and also, in a steric configuration in which the two Gluc complementary fragments can fold and thus reconstitute catalytic activity. Gluc activity can also be reconstituted with virus-like particles that express surface S-protein with detectable signal over background within 5 min of incubation. Design principles presented here can be readily applied to develop reporters to virtually any protein with sufficient available structural details. Thus, our results present a general framework to develop reporter assays for COVID-19, and the strategy can be readily deployed in response to existing and future pathogenic threats and other diseases.Virtual reality (VR) is known to cause cybersickness, and studies report that deteriorations of postural stability coincides with the onset cybersickness. It is unclear whether these deteriorations are the cause or a consequence of cybersickness. Thus, it is also unclear whether measures of postural stability may either predict susceptibility (cause) or objectively measure (consequence) the malaise. To examine whether deteriorations of postural stability can either predict or objectively measure cybersickness, healthy active adults (n = 50) were exposed to one of two different 10 min 360˚ VR videos.
